The use of some traditional methods such as lactose fermentation and indole production are not recommended due to limitations with these tests.
For example, a percentage of total coliforms do not ferment lactose, or do so slowly and may provide a false negative result.
Similarly, a number of Escherichia coli strains do not produce indole whereas some strains of Klebsiella spp. do. These tests could also generate spurious results.
Biochemical profiling tests such as BBL Crystal, API 20NE and VITEK all have significant advantages over individual biochemical tests.
These tests still have limitations, such as the size of the profile library.
